Abstract
A complete amino acid sequence of protein was determined by mass spectrometry with the tentative human consensus sequence (THC) database. To evaluate the advantages of THC database, it was applied for identification and in silico complete amino acid sequence determination of a novel protein p150, the mammalian target of rapamycin (mTOR)-associated protein. It was observed that expressed sequence tag (EST) database is a good source for protein identification and characterization in proteomics, even in the absence of entry in protein sequence database. The results show a complete amino acid sequencing of p150 by assembling both sequences of THC597050 and KIAA1303 in silico, which was confirmed by PCR and d'-rapid amplification of cDNA ends.
