Effect of Sargassum echinocarpum extract on Cu-Zn superoxide dismutase expression in liver and kidney of diabetic rats

Abstract

Hyperglycemic can increase the level of free radicals and reduce the activity of antioxidant enzymes. Sargassum echinocarpum is one of the brown seaweeds which contains bioactive compounds with hypoglycemic and free radical scavenging activities. This study aimed to evaluate the administration of S. echinocarpum extract (SEE) on the expression of Cu, Zn-Superoxide dismutase in liver and kidney of diabetic rats. S. echinocarpum was obtained from Talango waters, Sumenep District. Male rats Sprague-Dawley strain weighing 125-150 g were used in this study. Streptozotocin was used as a hyperglycemic inducer. This research was designed to five groups: normal, diabetic, diabetic + 150 mg/kg of SEE, diabetic + 300 mg/kg of SEE, and diabetic + 450 mg/kg of SEE, respectively, and all of the groups were treated for 90 days. The expression of Cu, Zn-Superoxide was observed by immunohistochemistry method. The results showed that 450 mg/kg of SEE is the strongest doses to decrease the blood glucose level and preserve the expression of Cu, Zn-Superoxide dismutase in the liver and kidney of diabetic rats. In conclusion administration of 450 mg/kg of SEE able to decrease the blood glucose level and reserve the Cu, Zn-Superoxide dismutase and this substance can be generated as an anti-stress oxidative agent on diabetes.
