Effect of Centella asiatica to developmental process of lead-induced zebrafish larvae

Abstract

Lead (Pb) is one of the heavy metals which is harmful to the environment. Its toxicity due to the oxidative stress occur. Most lead studies focus on neurotoxic effects but little research directed at other tissues that affect the disruption of embryonic development. Centella asiatica (CA) has a nutrient content that acts as an antioxidant to balance oxidants in the cells so as to prevent oxidative stress. Besides C. asiatica can prevent disruption of embryonic development in the stage of proliferation and differentiation. The aim of this study is to evaluate the effect of CA extracts to prevent developmental disorders in Lead-induced zebrafish larvae. Zebrafish (D. rerio) embryos were exposed to Pb with concentration of 2,5 ppm and CA concentration of 1.25, 2.5, and 5 μg/mL. This study observed the survival rate, hatching rate, mortality rate, body length, heart rate, locomotor activity, and zebrafish larvae bone ossification during the development stage. The results showed that CA decreased mortality rate, increases hatching rate, and significantly decreases heart rate (p <0.05), increases body length, bone ossification (p <0.05) and motility in Pb-induced zebrafish larvae. So that it can be concluded that Centella asiatica can prevent developmental disorders in zebrafish larvae exposed to Pb.
